DMD Highlight: Histone H3 Lysine 4 Trimethylation, Lysine 27 Trimethylation, and Lysine 27 Acetylation Contribute to the Transcriptional Repression

In this study by Yu and coworkers, expression of the transporter SLC47A2 was found to be transcriptionally suppressed in renal cell carcinomas compared to adjacent healthy tissue. This was due to a decreased ratio of histone H3 Lysine 4 Trimethylation to Lysine 27 Trimethylation on the SLC47A2 gene. Through deacetylation of histone H3 Lysine 27, histone deacetylase 10 regulates the H3K4me3/H3K27me3 ratio and hence the expression of SLC47A2. Thus, SLC47A2 was identified as a potential biomarker for renal cell carcinoma, which could help in early diagnosis of the disease.
