DMD Highlight: Implications for Metabolite Quantification by Mass Spectrometry in the Absence of Authentic Standards

Although quantification of metabolites by mass spectrometry in the absence of authentic reference standards or without a radiolabel is known not to be truly quantitative, but the caveats of quantifying metabolites based on parent drug response are not well appreciated.  Analysis of 71 clinically relevant drug and metabolites revealed that the ESI response of metabolites compared to their parent drugs ranged from 70 fold lower to 9-fold higher.  This highlights that the practice of using parent drug response to quantify metabolites should be used with caution.
