DMD Highlight: Effect of Disease-Related Changes in Plasma Albumin on the Pharmacokinetics of Naproxen in Male and Female Arthritic Rats

DMD May Highlights Li et al 

Li, Dubois Almon and Jusko assessed the impact of arthritis-related changes in plasma albumin on naproxen pharmacokinetics in male and female rats.  They found that saturable albumin binding accounts for the nonlinearity of naproxen PK, as well as part of the PK differences in arthritic rats. Reduced albumin concentrations, reduced overall drug binding and reduced naproxen clearances in arthritis led to elevation of the Cmax and area under the plasma concentration-time curve (AUC) of unbound drug.
