Research Areas

My long-term goal is to develop non-invasive approaches to predict variability in drug disposition by better understanding the genetic and non-genetic mechanisms of inter-individual differences in drug metabolism, transport and pharmacokinetics. Current research in my laboratory focusses on the variability caused by drug transporters and non-cytochrome P450 enzymes. We quantify transporters, enzymes, and their endogenous substrates in human tissues and biofluids by quantitative proteomics and metabolomics approaches, and then integrate these data along with genomics information into physiologically based pharmacokinetic (PBPK) models. Such integrated approach also allows identification and validation of surrogate biomarkers of drug transporters, metabolizing enzymes, and receptors, which are particularly important for the non-invasive prediction of drug disposition and response in special populations (e.g., children, elderly, and pregnant women) in whom clinical trials are not allowed.
